A simple mathematical model to determine the ideal empirical antibiotic therapy for bacteremic patients BJID
Tuon,Felipe F.; Rocha,Jaime L.; Leite,Talita M.; Dias,Camila.
BACKGROUND: Local epidemiological data are always helpful when choosing the best antibiotic regimen, but it is more complex than it seems as it may require the analysis of multiple combinations. The aim of this study was to demonstrate a simplified mathematical calculation to determine the most appropriate antibiotic combination in a scenario where monotherapy is doomed to failure. METHODS: The susceptibility pattern of 11 antibiotics from 216 positive blood cultures from January 2012 to January 2013 was analyzed based on local policy. The length of hospitalization before bacteremia and the unit (ward or intensive care unit) were the analyzed variables. Bacteremia was classified as early, intermediate or late. The antibiotics were combined according to the...

Discovery after discovery, host-associated microbiota reveal a growing list of positive effects on host homeostasis by contributing to host nutrition, improving hosts’ immune systems and protecting hosts against pathogens. In that context, a collection of oyster associated bacteria producing antibacterial compounds have been established to evaluate their role in non-host-derived immunity. Here, we described alterins; potent anti-Gram negative compounds produced by Pseudoalteromonas hCg-6 and hCg-42 isolated from different healthy oyster hemolymph. The strains hCg-6 and hCg-42 produce a set of at least seven antibacterial compounds, ranging from 926 to 982 Da structurally characterized as cyclolipopeptides (CLPs). Alterins share the same cationic...

To contribute to the development of antibacterial products from propolis produced by native Brazilian bees, twenty-nine samples of propolis collected from hives in the state of Minas Gerais, Brazil, were screened for in vitro activity against Aeromonas hydrophila, Bacillus subtilis, Pseudomonas aeruginosa, and Staphylococcus aureus. Among the samples from native Brazilian bees, only that from Frieseomelitta varia (Lepeletier, 1836) inhibited in vitro bacterial growth. Consequently, this propolis underwent fractionation by chromatographic methods monitored through Agar-diffusion assays with these bacteria, which resulted in the isolation and identification of 3,5-diprenyl-4-hydroxycinnamic acid (artepillin C), which showed MIC of 62.5 and 250 µg/mL against...

Antimicrobial susceptibility of Clostridium perfringens isolated from piglets with or without diarrhea in Brazil BJM
Salvarani,Felipe Masiero; Silva,Rodrigo Otávio Silveira; Pires,Prhiscylla Sadanã; Cruz Júnior,Eduardo Coulaud da Costa; Albefaro,Isabella Silva; Guedes,Roberto Maurício de Carvalho; Lobato,Francisco Carlos Faria.
The minimum inhibitory concentration (MIC) was determined for 13 antibiotics against Clostridium perfringens isolated from Brazilian piglets. The collection of isolates was performed in June to October 2010. All isolates were susceptible to amoxicillin and ceftiofur, whereas most were resistant to tetracycline and lincomycin. Avilamycin and narasin were more effective against isolates from non-diarrheic than from diarrheic piglets. The other antimicrobials were less active in need of high concentrations to inhibit the growth of the C. perfringens type A. These results suggest the need for further studies evaluating molecular factors related to the antimicrobial resistance of C. perfringens.

Hassi,Mohammed; El Guendouzi,Souraya; Haggoud,Abdelleatif; David,Susana; Ibnsouda,Saad; Houari,Abdellah; Iraqui,Mohammed.
The treatment of tuberculosis has become more difficult with the worldwide spread of multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains of Mycobacterium tuberculosis. Moreover, the prevalence of human disease caused by atypical mycobacteria has also increased in the past two decades and has further complicated the problem of the treatment of mycobacterial infections. It is therefore urgent to develop new highly active molecules against these bacteria. The present study reports the isolation from a Moroccan soil of a Bacillus strain that exhibits an important antimycobacterial activity. The strain was identified as Brevibacillus laterosporus using DNA sequencing of the 16S ribosomal RNA gene. The antimycobacterial activity was assigned...

Application of the antibiotic disk diffusion method to multivariate profiling of soil bacterial community: comparing the power to discriminate different soils and dimension of the discrimination with that of the Biolog method BJM
Doi,Ryoichi.
The antibiotic disk diffution (ADD) method was compared with the Biolog method in terms of power to discriminate soils and dimension of the discrimination. Soils from a forest and a citrus field in Thailand were profiled with these methods. These methods differentiated the soils in the principal component score plots. Then, Wilk's lambda statistic was determined to estimate power of these methods to discriminate the soils. The ADD method scored Wilk's lambda of 0.003 (p = 0.144) and 0.000 (p=0.020), for direct and ratio-transformed calculation, respectively. The Biolog method recorded Wilk's lambda of 0.001 (p=0.067), 0.003 (p=0.144) and 0.035 (p=0.440), at 0.25, 0.50 and 0.75 average well color developments (AWCDs), respectively. Zilli,Jerri Édson; Valisheski,Romano Roberto; Freire Filho,Francisco Rodrigues; Neves,Maria Cristina Prata; Rumjanek,Norma Gouvêa.
In order to contribute for the optimization of biological nitrogen fixation (BNF) associated with cowpea in Cerrado areas in the Northeast region of Brazil, this work aimed to analyze the diversity of rhizobial populations in eight areas of Cerrado, during a soybean and rice-cowpea rotation. Morphological traits (mucous production and colony morphology), genotypic analyzes (ARDRA 16S) and intrinsic resistance to antibiotics were determined for a collection of isolates captured using cowpea as a host-plant. The morphological data showed a inverse correlation (p < 0.05) between the number of legume (soybean and cowpea) crops, according to the history of each area, and rhizobium diversity, estimated by the Shannon-Weaver index. ARDRA data showed that...

Bacteriocins: exploring alternatives to antibiotics in mastitis treatment BJM
Pieterse,Reneé; Todorov,Svetoslav D..
Mastitis is considered to be the most costly disease affecting the dairy industry. Management strategies involve the extensive use of antibiotics to treat and prevent this disease. Prophylactic dosages of antibiotics used in mastitis control programmes could select for strains with resistance to antibiotics. In addition, a strong drive towards reducing antibiotic residues in animal food products has lead to research in finding alternative antimicrobial agents. In this review we have focus on the pathogenesis of the mastitis in dairy cows, existing antibiotic treatments and possible alternative for application of bacteriocins from lactic acid bacteria in the treatment and prevention of this disease.

Conjunctival endogenous microbiota in patients submitted to cataract surgery BJM
Locatelli,Claudete I.; Kwitko,Sérgio; Simonetti,Amauri Braga.
Bacterial isolation, identification and antimicrobial susceptibility tests were carried out in ocular material collected with swab and polimethylmethacrylate (PMMA) or silicone intraocular lenses (IOL) from forty six patients submitted to cataract surgery. Seventy six isolates and seven different microorganisms were identified. Coagulase-negative staphylococci (CNS) were the predominant microorganisms isolated from swabs (71.4% of cases), PMMA lenses (81.3%) and silicon lenses (77.8%). Coagulase-negative staphylococci isolates revealed high resistance to penicillin G followed by tetracycline, chloramphenicol and aminoglicosides. However, these isolates displayed great susceptibility to vancomycin, cephalothin and ofloxacin. Except for penicillin G,...

Economic Impact of a Ban on the Use of Over-the-Counter Antibiotics in U.S. Swine Rations AgEcon
Hayes, Dermot J.; Jensen, Helen H.; Backstrom, Lennart; Fabiosa, Jacinto F..
Antibiotic drugs are currently used in 90 percent of starter feeds, 75 percent of grower feeds, more than 50 percent of finishing feeds and at least 20 percent of sow feeds (USDA/APHIS). A ban on the use of feed-grade antibiotics would lead to changes in production processes and practices in production of pork, and hence would have an economic impact on the U.S. pork industry and pork market. This study considers the economic effects of a ban in pork production, with no change of regulation on other meats. The analysis uses a set of technical assumptions that are based in large part on a historical analysis of how the Swedish ban influenced the Swedish pork industry to anticipate the potential economic effects. A range of cases is examined. The cases use...

Effects on phytoplankton growth of dissolved substances produced by fish farming ArchiMer
Arzul, Genevieve; Clement, Alejandro; Pinier, Anne.
Fish Farming enriches the water column in dissolved organic and inorganic material. These materials excreed by the fish, elutriate from uneaten food and derive from other polluting substances. Ratios of the organic/inorganic forms of substances (for example N and P) are thus modified. Such perturbations in turn affect phytoplankton development. From in vitro analyses and tests of water taken from a turbot rearing tank, we show the changes in seawater chemical composition produced as well as the way growth was modified in three phytoplankton species: the diatom, Chaetoceros gracile, and the dinoflagellates, Gymnodinium cf. nagasakiense and Alexandrium minutum. Elutriates from the granules used for feed (0.1 to 100 mg.l(-1)) were found to be stimulating,...

Employing pharmacokinetic and pharmacodynamic principles to optimize antimicrobial treatment in the face of emerging resistance BJM
Santos Filho,Lauro; Kuti,Joseph L.; Nicolau,David P..
Antimicrobial efficacy in vivo is not exclusively defined by the activity of an antibiotic as determined in the in vitro susceptibility test. Knowledge of the pharmacokinetics and pharmacodynamics of antimicrobials and all phenomena occurring between antimicrobial agents and microorganisms is imperative. The pharmacodynamic (PD) parameters most often used in studies of antibiotic effect include the following relationships: the maximum free concentration (fCmax) to minimum inhibitory concentration (MIC) ratio, the free area under the curve (fAUC/MIC) ratio and the duration of time the free concentration exceeds the MIC (fT>MIC). Utilization of known pharmacokinetic/ pharmacodynamic surrogate relationships should help to optimize treatment outcome,...
